Jimi Hendrix and SRV immediately come to mind when I think Strat.

Clapton and Jimmie Vaughan were such huge influences on me and are faves but I haven't spent enough time listening to so many of the players mentioned here.

I agree, but there is an underdog out there that really sings:

Anson Funderburgh (and the Rockets)

Edge. I know he does not have the chops of most of the players mentioned but he writes great songs and clearly knows how to creatively use the technology to come up with so e great tones.

In no particular order...

Ike Turner
Otis Rush
Buddy Guy
Buddy Holly
Bobby Fuller
Jimmie Vaughan
Stevie Ray
Anson Funderburgh
Ronnie Earl
Paul Size (The Red Devils)
Hollywood Fats
Kid Ramos
Johnny "Guitar" Watson
Junior Watson
Mike Bloomfield (he played one towards the end of his career)

Maybe you haven't heard of Paul Size, he was total badass in the Devils. They only made one record, "King, King" and it's a classic.

Jimi Hendrix
Ritchie Blackmore
David Gilmour
John Frusciante

Don't think anybody mentioned Eddie Hazel yet.

I put funkadelic in my post, as I like all of their guitarists.

We got to #24 before Gilmore was mentioned?!?!!?

I'd completely forgotten about him :##

Another of favourite strat players is little known outside of Australia, Ian Moss (ex-Cold Chisel).

Funny Clapton not mentioned.

I was just about to post my list -- can't go wrong with the classics:

Hendrix
Clapton
David Gilmour
Mark Knopfler

And in a more soul-oriented vein, Curtis Mayfield and Nile Rodgers.

In the early years of ZZ, Billy certainly had some great strat tone's..

I like most of the ones mentioned in the prior replies, but if I had to pick three, I'd say Jimi, which was one of my very influences when I first picked up the electric guitar, Clapton and Gilmour.
